Yahoo Stores is a great place to start. Just make sure you do your product research, register your business, and I recommend using a different merchant service than the one that comes with Yahoo. Advanced Credit Systems is really good. You can reach them at http://www.acs-cvp.com.
After your site is setup you can advertise through banner ads, Adwords, Search Engine Optimization (SEO) or any number of things. For more info on SEO try out the Digitalpoint forums (forums.digitalpoint.com) or SeoBook.com.
